An eyepiece with a reticle is used to determine the size of samples and their structures in linear units (millimeters or microns) measure the areas of structures in the sample and conduct the comparative analysis of particle counts. The reticle is placed in the plane of the field diaphragm with a division of 0.1mm. The eyepiece with a reticle is installed instead of a standard 10x eyepiece. A paired eyepiece without a reticle is installed in the second eyepiece tube.
The eyepiece is compatible with microscopes that have a 30mm eyepiece tube.
Setting up
Before taking measurements the eyepiece reticle must be calibrated for each objective that will be used. It requires a calibration slide – a stage micrometer with a scale. The minimum division is 0.01mm. The slide should be placed on the stage and the microscope should be focused on the scale. The eyepiece should be rotated in the eyepiece tube until the scales of a slide and reticle are aligned parallel to each other.
Calculation of the reticle value for each objective
If you use low-magnification objectives you should calculate how many divisions of the eyepiece reticle fit into the slide scale. If you use medium and high-magnification objectives you should calculate how many divisions of the slide fit into the eyepiece reticle division.
Calculation for each objective is carried out according to the formula: Е = ТL/A where E – reticle division value Т – stage division value specified on the stage micrometer (0.01mm) L – number of stage micrometer divisions A – number of reticle divisions.
The resulting values of the reticle division for each objective shall be recorded.
Calculating the true linear size of an object
When measuring an object first you should count the number of eyepiece reticle divisions that the object covers. The result is multiplied by the reticle value previously calculated for the selected objective. The resulting value will be the true linear value of the object.
Calculating the area of an object
To determine the area of the object structure the number of reticle squares that the structure covers is counted. If the structure covers only a part of the square it should be determined in advance as to how this square should be considered in the final calculation: ignored or added to the result. The eyepiece with a reticle can only be used for approximate evaluation of the area; for accurate calculations image analysis software is recommended.
Particle counting
The eyepiece with a reticle can be also used for the comparative counting of particles in the given area. First the number of small particles in a square is counted and multiplied by the number of squares. Then the number of large particles in the entire area is counted. The resulting counts are compared.
